Riproduzione asessuata e variabilità genetica La riproduzione nei batteri avviene tramite scissione binaria: prima di ogni divisione, il DNA batterico viene duplicato e ciascuna cellula figlia riceve una copia completa del patrimonio genetico. Specializzata: solo una porzione di DNA batterico adiacente al punto di inserimento del fago è trasferita alla nuova cellula. Questo accade quando il fago si “stacca” in modo impreciso dal cromosoma del batterio donatore. Plasmidi e coniugazione Molti batteri possiedono, oltre al cromosoma principale, piccole molecole di DNA circolare dette plasmidi, che contengono pochi geni spesso legati a caratteristiche utili (ad esempio, la resistenza a un antibiotico). I plasmidi si replicano autonomamente e possono, in alcuni casi, integrarsi nel cromosoma o trasferirsi con facilità da un batterio all’altro. Il plasmide F e il processo di coniugazione Un plasmide noto è il plasmide F (F come “fertilità”) di Escherichia coli. Esso permette la coniugazione, un meccanismo nel quale il DNA plasmidico viene trasferito a un batterio privo di plasmide attraverso un ponte citoplasmatico formato da speciali appendici chiamate pili coniugativi. I batteri che possiedono il plasmide F sono detti F+. I batteri che ne sono sprovvisti sono detti F-. Durante la coniugazione: La cellula F+ duplica il proprio plasmide F. Una copia è trasferita alla cellula F- attraverso il pilo coniugativo. La cellula F- diventa così F+, acquisendo la capacità di dare inizio a sua volta a nuove coniugazioni. Cellule Hfr e trasferimento del cromosoma Talvolta, il plasmide F si integra nel cromosoma batterico, trasformando la cellula F+ in una cellula Hfr (High frequency of recombination). In questi casi, durante la coniugazione, possono essere trasferite anche porzioni del cromosoma dell’ospite, che arrivano nella cellula ricevente e si ricombinano con il DNA già presente. Ciò genera combinazioni genetiche aggiuntive e contribuisce alla variabilità della popolazione batterica. Nuovo esempio di coniugazione In alcuni esperimenti, un plasmide che conferisce la capacità di metabolizzare un particolare zucchero è stato rilevato in batteri che non lo possedevano in precedenza.
